JWH-210 is a potent synthetic cannabinoid of the naphthoylindole family. Developed by Dr. John W. Huffman (hence the “JWH” prefix), it was originally created for research into the endocannabinoid system but transitioned into the illicit “Spice” or “K2” market due to its high affinity for cannabinoid receptors.

As of 2026, it is considered one of the most potent members of the “first-generation” synthetic cannabinoids.

Chemical Profile & Potency

JWH-210 acts as a full agonist at both the CB1 and CB2 receptors.

• Binding Affinity: It has an exceptionally high affinity for the CB1 receptor ($K_i$ of 0.46 nM), making it significantly more potent than natural THC, which is only a partial agonist

• Potency Comparison: It is roughly 5 to 10 times more potent than JWH-018 and much stronger than natural cannabis.

• Structure: It is a 4-ethylnaphthalen-1-yl analog. The addition of the ethyl group increases its lipophilicity, allowing it to cross the blood-brain barrier rapidly and remain in fatty tissues for extended periods.
